ELISA Kit Operating Precautions:
- Reagents should be stored according to the instructions on the label and allowed to return to room temperature before use. Diluted standards must be discarded immediately and cannot be stored.
- Unused microtiter plates should be returned to the sealed bag promptly to prevent degradation.
- Unused reagents should be properly covered or sealed. Do not mix reagents from different batches, and always check the expiration date before use.
- Use disposable tips to avoid cross-contamination. Avoid using pipettes with metal parts when handling stop solution and substrate A and B.
- Prepare wash solution in clean plastic containers. Ensure all kit components and samples are well mixed before use.
- After washing the microplate, pat it dry thoroughly. Do not place absorbent paper directly into the wells to remove moisture.
- Substrate A should be kept tightly sealed to avoid evaporation. Substrate B is light-sensitive and should be protected from prolonged exposure. It is toxic, so handle with care. Read OD values immediately after completing the experiment.
- Ensure the reagent addition order is consistent across all wells to maintain uniform incubation time.
- Follow the incubation time, reagent volume, and sequence as specified in the protocol.
ELISA Kit Performance:
- Sensitivity: The minimum detectable concentration is lower than the first standard. The dilution curve shows good linearity, with a correlation coefficient (R) of 0.990 between sample results and expected concentrations.
- Specificity: The kit does not cross-react with other cytokines.
- Repeatability: The coefficient of variation (CV) between plates is less than 10%.
